Under the conditions of this bioassay, 4,4'-thiodianiline was carcinogenic for Fischer rats, inducing tumors in the liver, thyroid, colon, and ear canal of male rats, and the thyroid, uterus, and the ear canal of female rats. 4,4'-Thiodianiline was carcinogenic for B6C3F1 mice, inducing tumors in the liver and thyroid of both males and females.

4,4'-Thiodianiline causes most damage in the liver and thyroid. In long-term feeding studies, metastasizing tumours were found in the liver and thyroid and also in the uterus and mammary glands of rats and mice.

After short-term oral administration to pregnant rats, 4,4'-thiodianiline reduced the number of implantations. The compounds 4,4′-sulfonyldianiline (dapsone) and 4,4′-thiodianiline (I) were tested for possible carcinogenicity in F rats and B 6 C 3 F 1 mice.

Groups of 35 rats and 35 mice of each sex were given either dapsone or compound (I) in the feed. Matched controls consisted of 15 untreated rats and 14 untreated mice of each sex.
